The Axon Initial Segment (AIS) is a unique compartment at the base of axons. The AIS generates and shapes the action potential before it spreads along the axon. Thus, neuronal excitability depends crucially on the composition and position of the AIS, and these adapt to developmental and physiological conditions. Ankyrin G, also known as ankyrin-3, is a membrane cytoskeletal linker. It may be involved in the targeting and clustering of ion channels and cell adhesion molecules at the nodes of the Ranvier and axonal initial segments (AIS). It is therefore very often used as a marker for AIS.
